WebOct 5, 2024 · After auto-picking in RELION, the low-quality images and false-positive particles were removed manually. About 275k particles were extracted for subsequent 2D and 3D classification. We used the cryo-EM structure of human PTCH1* (EMD-7795) low-pass filtered to 60 Å as the initial model for 3D classification in RELION. WebMask one of the halfmaps to include only the central hexamer in Dynamo then use your favourite program and determine an appropriate threshold for binarisation. Use this as the --ini_threshold parameter in the following relion_mask_create command, replacing as appropriate:

WebOct 5, 2024 · Hi Oli, Thanks for the maps. Running the provided command with relion 2.1.b1 and the maps I don't believe I was able to reproduce the issue. Rather I've got the expected results out, a map that encloses the region specified by the threshold with the extended voxels and a soft edge.
